The ability to readily monitor metabolite concentrations in cell culture media could enable tighter culture condition control and potential improvements in yield and clinical potency of a cell therapy product. In this work, a microfluidic autosampler for in vitro measurements of glucose using Raman spectroscopy for bioprocessing applications is demonstrated. The platform aseptically delivers samples from multiple input vessels to a microfluidic optical flow cell. Discrete sample plugs prevent cross-talk between measurements and minimize dead volume. Raman spectroscopy enables glucose measurements without reagent consumption with a linear range from 0.5 to 4mg/mL. Automated measurements of glucose levels in mesenchymal stromal cell (MSC) culture media demonstrate a limit-of-detection (LOD) of 0.58 mg/mL – sufficient for MSC and other cell culture glucose monitoring.
